  • Histologically confirmed intracranial malignant glioma (GBM, anaplastic astrocytoma, anaplastic oligodendroglioma, anaplastic mixed oligoastrocytoma, malignant astrocytoma NOS) or histologically/radiographically confirmed recurrent or progressive meningioma
  • Progressive disease or tumor recurrence on imaging
  • Phase I: ≤3 prior relapses and ≤2 prior chemotherapy or biologic therapy regimens; Phase II: ≤2 prior relapses and ≤2 prior chemotherapy or biologic therapy regimens
  • Prior radiotherapy completed ≥4 weeks ago for progressive disease; patients with GBM without progression after external beam radiotherapy are eligible; progression 4–12 weeks post-radiotherapy requires clear MRI evidence

  • No active infection
  • No other serious concurrent medical illness
  • No other malignancy within past 3 years except nonmelanoma skin cancer or carcinoma in situ of cervix
  • No disease that would obscure toxicity or dangerously alter drug metabolism
