  • Confirmed diagnosis of NF2 by NIH or Manchester criteria or NF2 gene mutation detection
  • Progressive or symptomatic meningioma with ≥20% volume increase or ≥3 mm growth over past 2 years
  • Target meningioma not amenable to surgery due to patient preference or high surgical risk
  • Measurable disease: at least one meningioma ≥1.0 ml on contrast-enhanced MRI within 28 days of registration
Key exclusion· 14
  • Prior chemotherapy, biological therapy, or radiation within 21 days (investigational monoclonal antibody within 3 months; nitrosoureas/mitomycin C within 6 weeks)
  • Unresolved Grade >1 toxicities (except alopecia) from prior anti-tumor treatment
  • Major surgery within 4 weeks or minor surgery within 14 days before first dose (excluding tumor biopsies)
  • Prior meningioma progression during mTORC1/2 inhibitor treatment (but not mTORC1 inhibitors like everolimus)
